Training Day Background


The training day aims to divulge the use of standardise protocols to facilitate future polyP research. While the main techniques associated to polyP research do not require sophisticate instrumentations, published protocols often lack important details. The training day aims at addressing this issue by providing practical demonstrations and training in the main biochemical and cell biology methods associated with polyP research.

The training day objective is to demonstrate the following biochemical and cell biology methods associated with polyP research.

 

Topics:

  • polyP extraction and purification procedures
  • polyP enzymology; the purification and use of recombinant His-EcPPK, His-ScPPX1 and His-ScDDP1
  • analysis of polyP by gel electrophoresis and visualization by Toulidine Blue and DAPI staining
  • Quantification of polyP by DAPI or JC-D8 fluorescence and Malachite green assay.
  • Microscopy detection of polyP.

Each section will be carefully discussed and critical steps will be outlined and directly demonstrated.
